Vascular dementia is the second leading cause of dementia. The molecular mechanisms of human vascular dementia are not understood. Vascular dementia occurs in the cerebral white matter, and is centered around blood vessels, astrocytes and their associated cells. This is termed the “neurovascular unit”: endothelial cells, astrocytes, pericytes, oligodendrocyte progenitor cells and possibly perivascular fibroblasts. This Post-Doctoral fellowship or Project Scientist position will lead a project to identity the molecular changes in these cells of the neurovascular unit in humans with aging and with vascular dementia. The techniques will use human Nuc-seq from autopsy studies. This study is a close collaboration of several labs at UCLA, including Drs. S. Thomas Carmichael, Inma Cobos, William Lowry, Luisa Iruela-Arispe and Ben Novitch. The position will work between the labs and benefit from a rich scientific environment, with expertise in vascular dementia and neuronal, vascular and glial biology. Nuc-seq will be performed on carefully selected human white matter samples, RNAseq performed from these samples, and bioinformatics analyses—all done in a pipeline that we have already established. A PhD and Molecular biological experience are required for this position. This project will identify human gene systems that are abnormal in these neurovascular cell types, during aging and vascular dementia. These genes systems will then be tested for their mechanistic roles.
